Requirements

Expertise in functional genomics and genome engineering, including high-throughput CRISPR/Cas9, CRISPRi/a screens, STARR-seq, and massively parallel reporter assays (MPRA)
Experience with single-cell and spatial genomics protocols, such as scRNA-seq, scATAC-seq, scCUT&Tag, spatial transcriptomics, and multimodal platforms (e.g., 10x Genomics Multiome, Visium)
Knowledge of transcriptional regulation techniques, including scGRO-seq, SLAM-seq, and nascent RNA profiling
Proficiency in epigenome editing tools like dCas9-p300 and dCas9-TET1 for modulating enhancer activity
Skills in primary immune cell manipulation, including isolation, culture, and genetic reprogramming of T cells, B cells, NK cells, dendritic cells, macrophages, and HSPCs
Ability to design synthetic biology circuits, such as logic-gated and inducible systems for controlled gene expression
Experience with ex vivo and organoid models, particularly engineering immune cells in human immune organoids
Background in integrating advanced functional genomics, single-cell multi-omics, synthetic biology, immunology, and bioengineering
High motivation to lead pioneering research in non-coding regulatory elements governing immune cell gene expression

Responsibilities

Apply high-throughput CRISPR/Cas9 and CRISPRi/a screens, STARR-seq, and MPRA to functionally map immune-cell-specific enhancers, silencers, and boundary elements
Develop and optimize protocols for scRNA-seq, scATAC-seq, scCUT&Tag, spatial transcriptomics, and multimodal platforms
Use scGRO-seq, SLAM-seq, and nascent RNA profiling to dissect transcriptional kinetics in engineered immune cells
Employ targeted histone modification and DNA methylation tools (e.g., dCas9-p300, dCas9-TET1) to modulate enhancer activity in immune contexts
Isolate, culture, and genetically reprogram primary human and murine immune cells (T cells, B cells, NK cells, dendritic cells, macrophages, HSPCs)
Design logic-gated circuits and inducible systems for spatially and temporally controlled gene expression
Engineer immune cells within human immune ex vivo and organoid models
Lead research to identify and manipulate enhancers, silencers, and cis-regulatory elements controlling immune-modulating genes to reprogram immune responses
Integrate functional genomics, single-cell multi-omics, and synthetic biology to dissect immune regulatory circuits and apply strategies in in vivo models and human-derived systems
